President Cyril Ramaphosa said that the interception of the Global Sumud Flotilla in international waters is contrary to international law.
The Flotilla comprises of multiple vessels, with numerous international activists and aid workers, including South Africans, carrying humanitarian aid to the people of Gaza.
Some of the boats have been stopped by the Israeli navy overnight and some of those on board have been detained.
The Presidency said that efforts to obstruct such missions only deepen suffering and further isolate vulnerable communities.
Presidency Spokesperson Vincent Magwenya said, "The interception of the Global Sumud Fotilla is another grave offence, by Israeli PF, global solidarity and sentiment that is aimed at relieving suffering in Gaza and advancing peace in the region.
South Africa has reiterated its position that the blockade of Gaza is unsustainable and incompatible with international law.
The country continues to support calls made by the United Nations and other multilateral institutions urging the free and safe passage of humanitarian relief.
"On behalf of the government and the nation, President Ramaphosa calls on Israel to immediately release the South Africans abducted in international waters, and to release other nationals who tried to reach Gaza with humanitarian aid.
