Mobility MEC Isaac Sileku said he took the decision to protect lives after at least 10 people were killed in taxi-related violence in recent weeks.
The attacks are linked to a feud between rival taxi associations CODETA and CATA over the closed routes.
CODETA's Edgar Mohale said Sileku's decision to close the routes was very unfortunate.
Mohale said there were no issues between his association and CATA.
"We are brothers; we are working together, so there's nothing between us and them. It's the government that is not giving us the right direction. They are just playing delaying tactics of closing the routes."
CODETA has hauled the provincial government to court to challenge the closures.
That matter will be back at the Western Cape High Court on Saturday.
