Police said they intercepted the Brazilian national shortly after landing, acting on intelligence information.
A search of the suspect’s luggage led police to uncover about 10kg of cocaine, with an estimated value of R4.2 million.
Police said the regular arrests of drug mules at the OR Tambo International Airport should send a stern warning that police were on alert to clamp down on criminality.
Spokesperson Amanda van Wyk: "SAPS continues to record commendable success in dismantling transactional drug syndicates operating between South Africa and other countries. Through targeted intelligence-led operations, more than 25 drug traffickers were arrested since the beginning of the year at this port of entry."
